Vitamin K2 Carboxylation: Osteocalcin and Matrix Gla Protein
Vitamin K does not add calcium to tissue. Instead, it supports an enzyme that changes selected protein residues, creating Gla sites with a stronger ability to coordinate calcium ions.
What is vitamin K-dependent gamma-carboxylation?
It is an enzyme reaction that adds a carboxyl group to selected glutamate residues, turning them into gamma-carboxyglutamate, or Gla.
Gla changes protein charge and geometry
The added carboxyl group provides extra negative charge. Multiple Gla sites can coordinate positively charged calcium ions and help the protein attach to mineral or membrane surfaces.
Vitamin K cycles during the reaction
Vitamin K hydroquinone supplies reducing power and becomes vitamin K epoxide. Recycling enzymes regenerate forms that can support additional carboxylation cycles.
What does osteocalcin do?
Osteocalcin is a protein produced by bone-forming cells, and its vitamin K-dependent Gla residues help it interact with calcium-rich hydroxyapatite.
The protein belongs to the bone matrix
Osteoblasts synthesize osteocalcin under signals that include vitamin D. Vitamin K then supports the post-translational step that changes its calcium-binding properties.
Production and activation are separate steps
Vitamin D-related gene signaling can increase a protein's production, while vitamin K supports its carboxylation. This explains why D3 and K2 are discussed together.
What is matrix Gla protein?
Matrix Gla protein is a vitamin K-dependent protein found in vascular smooth muscle, bone and cartilage, where its carboxylated form participates in calcium-related matrix regulation.
MGP is different from osteocalcin
Both contain Gla sites, but their tissue distribution and biological roles differ. One label term should not be used as a substitute for the other.
K2 complements mineral and botanical ingredients
In a D3 + K2 formula, these vitamins contribute calcium-regulatory signaling while beetroot, hawthorn and cayenne contribute nitrate, polyphenol and sensory pathways.

Frequently asked questions
What does Gla mean?
Gla means gamma-carboxyglutamate. It is a modified amino-acid residue created by a vitamin K-dependent enzyme.
Does vitamin K2 contain calcium?
No. It supports protein modification rather than supplying the mineral.
Are osteocalcin and MGP the same?
No. They are separate vitamin K-dependent proteins with different tissue patterns.
Why are D3 and K2 paired?
D3 supports expression of calcium-related proteins and K supports activation of selected proteins. Their functions intersect but remain distinct.
